abstract

to produce embryogenic callus and somatic embryos, tendril explants were collected and cultured in ms medium supplemented with various concentrations of hormones. after one month most of explants produced callus. the highest percent of embryogenic callus obtained in ms medium supplemented with 4.5 µm bap and 1µm 2,4-d. production of somatic embryos was depended on genotype and medium. yaghouti cultivar in ms medium without hormone, bidaneh sefid in ms medium supplemented with 10.4µm naa and 0.4µm bap and shahroudi cultivar in ms medium supplemented with 4.5µm bap and 1µm 2,4-d produced the highest percent of somatic embryos. for germination, embryos were transferred in to ms medium supplemented with 1µm bap at the torpedo stage. at this medium most of embryos germinated and produced plantlet .

Chloroplast genome diversity in Portuguese grapevine (Vitis vinifera L.) cultivars.

Grapevine chloroplast (cp) DNA diversity was analysed for the first time through amplification and digestion of fragments of the large single copy (LSC) region by polymerase chain reaction-restriction fragment length polymorphism methodology and also by amplification of three microsatellite loci, previously described as polymorphic in grapevine. Thirty-eight grapevine cultivars collected mainly...

grapevine (vitis vinifera l.) regeneration via somatic embryogenesis from whole flower explant

abstract to produce embryogenic callus and somatic embryo, whole flower explants were collected at two sampling stages of i, iii. to produce embryogenic callus and somatic embryo, ms medium supplemented with 5 and 10µm 2,4-d and 2µm bap were used. for embryo differentiation, ms medium with 0.5mg/l iba, ms medium without any plant growth regulators, ms medium with 2mg/l iba and 0.2mg/l bap, ms m...

Transcriptional Analysis of Tendril and Inflorescence Development in Grapevine (Vitis vinifera L.)

In grapevine (Vitis vinifera L.), the lateral meristem can give rise to either tendrils or inflorescences which are determined organs. To get insights into the processes of tendril and inflorescence development, we characterized the transcriptional variation taking place in both organs. The results of the global transcriptional analyses along tendril and inflorescence development suggested that...

Sequence Polymorphisms and Structural Variations among Four Grapevine (Vitis vinifera L.) Cultivars Representing Sardinian Agriculture

The genetic diversity among grapevine (Vitis vinifera L.) cultivars that underlies differences in agronomic performance and wine quality reflects the accumulation of single nucleotide polymorphisms (SNPs) and small indels as well as larger genomic variations. A combination of high throughput sequencing and mapping against the grapevine reference genome allows the creation of comprehensive seque...

Sugars and flowering in the grapevine (Vitis vinifera L.).

Sugars play an important role in grapevine flowering. This complex process from inflorescence initiation to fruit maturity takes two growing seasons. Currently, most of the available data concern the involvement of sugars as energy sources during the formation of reproductive structures from initiation of inflorescences during the summer of the first year, until flower opening during the follow...

Oxidative stress homeostasis in grapevine (Vitis vinifera L.)

Plants can maintain growth and reproductive success by sensing changes in the environment and reacting through mechanisms at molecular, cellular, physiological, and developmental levels. Each stress condition prompts a unique response although some overlap between the reactions to abiotic stress (drought, heat, cold, salt or high light) and to biotic stress (pathogens) does occur. A common feat...
